Simply, what moments in videogames have made you actually genuinely not-in-the-casual-Internet-sense laugh out loud? Slapstick, clever dialogue, or even so-bad-it's-hilarious moments. Have at it!
My opening contribution, perhaps unsurprisingly given how much I've been harping on about it recently, comes from the Phoenix Wright Trilogy. I've had a guilty chuckle at pop culture references (like Moe the clown parodying various TV shows, or innocent sidekick Maya slyly saying, "This one time, at lawyer camp, Nick..."), but today's hilarity comes from a moment in T&T, when the court are observing a witness' drawing:
Defence: "Are..." Prosecution: "You..." Judge: "High! The girl... She's really high up in this picture!"

Ah - I can't think of an example right now, typically, but at least half a dozen lines in Earthbound have caused me to giggle.
One example that's popped into my head: "Kidnapping is wrong! I'll be careful not to kidnap anyone."
I don't think you'll ever get this again. Some of the lines are genuinely funny but also slightly incongruous English; and I think the translation's got to the stage where the text will be more fluent, and therefore some of Earthbound's humour would disappear.

Bishi Bashi Special and Hogs of War had loads of them. There aren't too many games with scripted moments that made me laugh out loud though. Most of the hilarity from the likes of Super Monkey Ball, Wii Sports and the WWE games came from the multiplayer with my mates.

Most recent example I can think of is Grand Theft Auto V. There's an eccentric British couple called Nigel and Mrs Thornhill. They love chasing celebrities and collecting memorabilia from them, and the cutscenes involving them have me in near stitches. Here's your first meeting with them

It was a later meeting with them that cracked me up earlier, especially when a celebrity Nigel and Mrs Thornhill were chasing spotted them "Oh shit, it's that crazy English bitch again" he said "Nigel! He remembers me!" was Mrs Thornhill's response. Brilliant characters and some very funny cutscenes.

The one big laugh I'm remembering at the moment is the first thing I picked up in Dragon Quest IX being manure. "Simon acquires some horse manure!" The game seemed so happy that I was doing it I burst out laughing.

Anything that happened in house of the dead overkill was hilarious.
Also, my mate and I have been playing Digimon World on the PS1 and some of the awful translation work is great.
