(As usual, searched through the first four pages of GG, couldn't find a thread on this, made me own. Happy for this to be merged with another thread if someone digs one up.)
A simple question: what are they? A character has to actually say them, they can't be printed or narration like 'A WINNER IS YOU'. Here's a couple to start off:
'Thank you so much-a for to playing my game!'
No line so charming. Obviously this has all of the nostalgia in, but when you hear Mario chirp up with this upon finishing SM64, I defy you not to feel a little warmth in your withered charred heart.
No line so chilling. If you haven't realised by now that Majora's Mask is much darker than OoT...
'Your lives that I spit on are now but a caricature of a cartoon drawn by a kid who is stupid!'
No line so chortling. Fawful is a character almost made of one-liners, but this is my new favourite. I'm not usually an aggressive person, but I definitely want to use this insult on somebody.
Over to y'all!

Oh boy. I want to add to this, but I'm having a blank. There's a couple of good'ns in Earthbound, I remember from AGDQ:
Kidnapping is wrong. I’ll be careful not to kidnap anyone! - a resident in Twoson
It could only cost you your life, and you got that for free! - a sailor preparing for a dangerous voyage
Not doing very well here...

I don't know what the single best one-liner in gaming is, but it's almost definitely somewhere in this clip:
Especially for 8:22 onwards. I could type some of the lines out, but it's Rik Mayall's utterly exceptional vocal performance that really makes them great.
What a fucking great game, and what a fucking great man.

[absoluteham]"What is a man?" *throws glass of wine/blood* "A miserable little pile of secrets! But enough talk... have at you!"[/absoluteham]
That whole cutscene warrants a watch.

Whenever I'm asked a question like this, my mind goes blank. However, after many days of consideration, I have come up with two lines.
1) "May the way of the Hero lead to the Triforce." This repeated line comes from A Link To The Past. Each of the seven maidens ends their infodump with that line; I like it because it almost feels like a prayer, or at least a line from a religious text. Given that the game features a priest in a church (OK, OK, 'Sanctuary'), it feels appropriate. It also serves to connect the Dark World temples together in a way that Zelda games had never managed before... and haven't very often since.
2) "Ready for adventure!"
DIGRESSION ALERT: My phone suggested "Ready for advice!". Few games have that line.
Captain Toad's charming enthusiasm, both as a game and as a character, is encapsulated with this little beaut - far better than "Mah moh!", at least. Watching his limited abilities still manage to conquer huge bosses and carefully planned levels is a joy. I am really keen on a sequel to CT:TT - you could say I'm "ready for adventure"!

How dare you Zero, "Mah moh!" is a chuffing blinder. Recant!
